To do more than 500 points of damage with a melee hit, you might need to change your skill selection.

Reset your skills, then add points to the Brawler skill tree (Pumping Iron and Berserker). Then select an appropriate melee weapon, like the Katana.

Go find the punching bag in Dragan's gym. A charged melee attack will do the most damage.
